Design, synthesis and biological evaluation of new bis(thiosemicarbazone) derivatives as potential targeted anticancer agents for non-small cell lung cancer
Thiosemicarbazonesrepresent an important class of ligandsfortargeted therapyof many types of cancer including non-small celllung cancer. In order to identify potential antitumoragentsfor targeted therapy oflung cancer, new bis(thiosemicarbazone) derivatives (1-11) were preparedviathe reaction of 1,4-phenylenebis(thiosemicarbazide) with 5-arylfurfurals. The cytotoxic effects of compounds 1-11on A549human lung adenocarcinoma and L929 mouse fibroblast cellswere investigated using MTT test. Compounds 1, 10and 11were the most potentanticancer agents in this series on A549 cell line with IC50values of 14.33±0.47 μg/mL, 11.67±2.49 μg/mL and16.67±5.56 μg/mL, respectivelycompared to cisplatin (IC50= 18.33±0.94μg/mL). Based on theirIC50values for L929 cell line, their anticancer activitieswere found to be selective.Moreover,flow cytometry-based analyseswere performed to examine their effects on apoptosis and mitochondrial membrane potential.The treatment of A549 cells with compounds 1, 10and 11 at IC50 concentrationsled to the induction of apoptosis along with mitochondrial membrane depolarization. In order to explore theirmodeof action,compounds 1, 10 and 11were evaluated for their inhibitory effects onCOX-1 and COX-2 in A549 cells. In particular, N,N'-(1,4-phenylene)bis(2-((5-(2,5-dichlorophenyl)furan-2-yl)methylene)hydrazine-1-carbothioamide) (10) was identified as a selectiveCOX-2 inhibitor(6.96% for COX-1 and 54.81% for COX-2). According to these results, compound10warrants further in vitroand in vivostudies as a potential targeted anticancer agentfor the management of non-small celllung cancer.

Bu çalışmada, Eskişehir'de dört farklı sosyo-kültürel grubu temsil eden 500 kişinin vitamin kullanım alışkanlıkları ve bu alışkanlıkları etkileyen faktörler çalışılmıştır. Araştırmaya katılan denekler tesadüfi örnekleme yöntemi ile seçilmiş ve konu ile ilgili soruları yanıtlamaları istenmiştir. Araştırmanın verileri, genel olarak ve genel içerisindeki gruplara göre değerlendirilerek, % oran olarak verilmiş ve grafik olarak gösterilmiştir. Bu çalışmanın sonuçları, çoğu kişinin vitaminleri sağlık ve belirli hastalıklara direnç kazanmak için kullandığını göstermektedir. Günlük yaşamdaki beslenme bozukluğu ve gıdalara karşı oluşan güvensizlik vitamin kullanımını etkileyen diğer faktörler olarak ortaya çıkmaktadır. Bu noktada, vitamin tercihleri ve bilgilerine bakıldığında sosyo-kültürel gruplar arasında belirgin bir farklılık olduğu açıktır.; In the present study, vitamin usage habits of five hundred people representing four different sociocultural groups in Eskişehir and the factors affecting these habits were studied.The participants of the research were chosen by random sampling method and were asked to answer the questionary. The data of the investigation were given in percentages and shown in graphs in the view of the evaluation in general and in subgroups.The results of this study indicate that most people use vitamins for wellness and gaining resistance to certain diseases. Nutrition disorder in daily life and distrust of food are other factors affecting vitamin usage of people. At this point, considering their preferences and their knowledge of vitamins, it is clear that there is a visible distinction between the socio-cultural groups.

Mevcut antikanser ajanlara karşı rezistans gelişimi, yeni antikanser ajanlar için bir araştırma alanı yaratmaktadır. Bununla birlikte, normal hücreler üzerinde etki göstermeden ya da en az etkiyle anormal hücrelerin çoğalmasını seçici olarak inhibe edebilecek yeni bir ajanın geliştirilmesi oldukça güçtür. Bu nedenle, kanser kemoterapisi medisinal kimyacılar için çok önemlidir ve çeşitli kanser tipleri üzerinde antikanser aktivite gösterme olasılığı bulunan yeni kemoterapötik ajanların geliştirilmesi üzerinde çalışmalar halen devam etmektedir. Bu noktadan hareketle, bu çalışmada bazı biskinoksalin türevleri sentezlenmiş ve HT-29 (kolon kanseri) ve MCF-7 (göğüs kanseri) hücre hatları üzerinde antikanser aktiviteleri için taranmıştır. Sentezlenen bileşiklerden bazıları (3e ve 3l) MCF-7 hücre hattı üzerinde önemli sitotoksisite ve DNA sentez inhibisyonu göstermiştir.; Resistance improvement against the existing anticancer agents creates a research area for new anticancer agents. Nevertheless, it is rather difficult to develop a new agent which can selectively inhibit the proliferation of abnormal cells with least or no affect on normal cells. Therefore, cancer chemotherapy is very important for medicinal chemists and the studies are still being carried on to develop new chemotherapeutic agents that are probable to indicate activity on various cancer types. Depending on this point, in this study some bisquinoxaline derivatives were synthesized and screened for their anticancer activity on HT-29 (colon carcinoma) and MCF-7(breast carcinoma) cell lines. Some of the synthesized compounds (3e and 3l) showed significant cytotoxicity and DNA synthesis inhibition on MCF-7 cell line.

Amaç: Bu çalışmada bazı yeni piperazin bileşiklerinin antikolinesteraz amaçlanmıştır. Gereç ve Yöntem: On yeni piperazin türevi sentezlenmiştir. Sentezlenen bileşiklerin yapı aydınlatmaları spektroskopik yöntemler ile gerçekleştirilmiştir. Bu bileşiklerin asetilkolin esteraz (AChE) ve butirilkolin esteraz enzimleri üzerindeki etkileri, Elmann kolorimetrik metodu ile belirlenmiştir. En etkili 2b bileşiği için Lineweaver-Burk grafiği kullanılarak enzim kinetik çalışmaları gerçekleştirilmiştir. Bulgular: Klor değişken grubu içeren 2b kodlu bileşiğin 1mM, 0.1 mM and 0.01 mM konsantrasyonlarda sırasıyla % 82.95, % 66.93 ve %42.63 inhibisyon oranları ile en etkili türev olduğu belirlenmiştir. Enzim kinetik çalışmaları sonucunda 2b kodlu bileşiğin non-kompetitif tip inhibisyon gösterdiği tespit edilmiştir. Sonuç: Hiçbir bileşik standart ilaçlar donepezil ve takrin kadar etki göstermemiştir. Ancak 2b ve 2c kodlu bileşikler AChE enzimine karşı seçici ve umut verici düzeyde inhibisyon sergilemiştir.; Purpose: The aim of present study is to evaluate anticholinesterase activities of some new piperazine compounds. Materials and Methods: Ten new piperazine derivatives were synthesized. Structure elucidation of the synthesized compounds was performed by spectroscopic medhods. Inhibitory activities of these compounds on acetylcholine esterase (AChE) and butrylcholine esterase (BChE) enzymes have been determined by Ellman's colorimetric assay. Enzyme kinetic studies were performed for the most active compound 2b by using Lineweaver-Burk plots. Results: The compound 2b including chloro substituent was found as the most active derivative with 82.95 %, 66.93 % and 42.63 % inhibition rates at 1 mM, 0.1 mM and 0.01 mM concentrations, respectively. Noncompetitive type of inhibition was determined for compound 2b as a result of enzyme kinetic studies. Conclusion: None of the compounds showed activity as much as standard drug donepezil. However, compound 2b and 2c displayed a promising and selective inhibitory activity against AChE.
