Anti-LDL antibody-nanoparticles embedded cryogel for low density lipoprotein-depletion from hypercholesterolemic human serum
Abstract
In this study, we have focused our attention on developing a novel immunoadsorbent while combining cryogel-based specific sorbents with immunoaffinity (anti-LDL antibody). A novel method has been developed by our group abbreviated as ANADOLUCA in which biomolecules could be converted into protein-like fluorescent nanoparticles through ruthenium-mediated polymerization and crosslinking reactions. Herein, anti-LDL antibody nanoparticles that have an average particle size as 50 nm with a narrow size distribution have been prepared via ANADOLUCA method. These nanoparticles were then embedded in the polymeric structure to give immunospecificity to the cryogens during the cryo-polymerization process under semi-frozen conditions. After characterization of the materials by scanning electron microscopy and swelling tests and optimization of depletion, conditions have been investigated by varying affecting parameters like initial LDL concentration and pH of the solution. Finally, the cryogel column developed has been conducted to fast protein liquid chromatography system as a stationary phase to evaluate its chromatographic performance not only from aqueous LDL solution and but also from human serum in a single step separation process. As a result, an efficient LDL depletion performance has been achieved for the cryogel column in short treatment time. Therefore, the cryogel column might be classified as potential cost-friendly and efficient candidate for immunoaffinity depletion of LDL molecules from hypercholesterolemic human serum.
