Prokaryotic diversity of the two solar salterns in Sungurlu were investigated by using culture-dependent techniques and molecular approaches including fluorescence in situ hybridization (FISH) and high-throughput sequencing. Total cell count using DAPI revealed the samples had approximately 10(6)cells/mL. Archaeal and bacterial cells were counted by using domain specific probes showed that the proportion of the archaeal cells outnumbered bacterial cells. The percentages of the archaeal reads obtained from Illumina MiSeq sequencing were relatively higher than that of the bacterial reads. Most of the reads were assigned to the family Halobacteriaceae and genus Salinibacter. Salicola, Salinibacter, Halovibrio, Halomonas Haloarcula, Halorubrum and Halobacterium related strains were obtained from the cultivation studies.en_US
